Abstract

This dissertation observes the effects of the steroid hormone, progesterone, on telomerase-immortalized human endometrial stromal fibroblasts in vitro, using a Luciferase assay. The methods leading up to the assay include doing a Midi-prep for plasmid isolation as well as plating, seeding (into a 96-well-plate), transfecting (with isolated plasmids), and exposing the fibroblasts to progesterone. The data produced was inconsistent across the three replicates, and the comparison ratios for Luciferase activity did not indicate any trends in progesterone dose response over increasing doses. However, when optimized, this project will be used as a baseline in the in vitro comparative system that our laboratory is developing to compare progesterone dose response between different ape endometrial stromal fibroblasts.
